Oligonucleotide-coupled Beads

Oligonucleotide-coupled beads are microscopic particles functionalized with short, sequence-defined DNA or RNA strands, creating solid supports for selective nucleic-acid capture and detection. Their oligonucleotides hybridize to complementary target sequences under controlled conditions, allowing bead-associated targets to be retained, enriched, or measured through a signal or downstream assay. In immunology and infection research, these beads support multiplexed analysis of pathogen genomes, antimicrobial-resistance markers, and immune-related transcripts, often by assigning distinct probe sequences to different bead populations. They can improve assay organization and specificity while reducing sample handling, helping researchers identify infectious agents and characterize host responses in complex biological samples.

Multiplex Detection of Bacteria in Complex Clinical and Environmental Samples using Oligonucleotide-coupled Fluorescent Microspheres

We describe a multiplex method for the detection of microorganisms within a sample using oligonucleotide-coupled fluorescent beads. Amplicon from all organisms within a sample is hybridized to a panel of probe-coupled beads. A Luminex or Bio-Plex instrument is used to query each bead for bead type and hybridization signal.

A protocol is presented for enriching host cell proteins (HCPs) from drug products (DP) and detecting peptides using proteome enrichment beads. The method is demonstrated using an in-house manufactured monoclonal antibody (mAb) drug substance (DS), which is a well-characterized reference material for evaluating and comparing different methods in terms of performance.

A Bead-Based Multiplex Immunoassay to Identify Pathogen-Specific Antibodies in Saliva

This video demonstrates a bead-based multiplex immunoassay technique to simultaneously detect antibodies for multiple environmental pathogens in human saliva. Fluorescent beads were coupled to antigens from different pathogens, which were captured using antibodies in the human saliva sample. Upon labeling the antibodies using a fluorescent reporter, the beads were analyzed using flow cytometry to assess the presence of different pathogen-specific antibodies in the saliva.
