Asc Sheet Transplantation

ASC sheet transplantation is a regenerative medicine technique that transfers sheets of adipose-derived stem cells (ASCs) to injured or diseased tissue, aiming to support repair while preserving the cells’ native organization. In this approach, ASCs are expanded under controlled culture conditions until they form contiguous layers rich in extracellular matrix; the sheets are then harvested and placed on the target site without fully dissociating the cells, helping retain cell-cell and cell-matrix interactions. Transplanted ASC sheets can provide paracrine signals and matrix components that promote tissue remodeling and healing. The method is being investigated for wound repair, musculoskeletal regeneration, and other cell-based therapies.

Adipose-derived stem cells (ASCs) are easily isolated and harvested from the fat of normal rats. ASC sheets can be created using cell-sheet engineering and can be transplanted into Zucker diabetic fatty rats exhibiting full-thickness skin defects with exposed bone and then covered with a bilayer of artificial skin.

This protocol focuses on utilizing the inherent ability of stem cells to take cue from their surrounding extracellular matrix and be induced to differentiate into multiple phenotypes. This methods manuscript extends our description and characterization of a model utilizing a bilayered hydrogel, composed of PEG-fibrin and collagen, to simultaneously co-differentiate adipose-derived stem cells1.

For creation of highly organized structures of complex tissue, one must assemble multiple material and cell types into an integrated composite. This combinatorial design incorporates organ-specific layered cell sheets with two distinct biologically-derived materials containing a strong fibrous matrix base, and endothelial cells for enhancing new vessels formation.
