Drug Sensitivity Testing

Drug Sensitivity Testing measures how biological cells, tissues, or infectious organisms respond to a medication, helping determine whether a treatment is likely to be effective. In a controlled laboratory assay, samples are exposed to defined drug concentrations, and outcomes such as cell viability, growth inhibition, or pathogen survival are compared to untreated controls; results may be summarized by values such as the half-maximal inhibitory concentration or minimum inhibitory concentration. In medicine, these tests support antimicrobial selection, cancer treatment planning, and investigation of drug resistance. They also provide evidence for personalized therapy and help researchers evaluate new drugs before clinical use.

We describe a high-throughput drug sensitivity assay for primary multiple myeloma cells. It consists of a reconstruction of the bone marrow microenvironment (including extracellular matrix and stroma) in multi-well plates, and a non-invasive method for longitudinal quantification of cell viability.

Determining the Role of Neutrophil-Like Cells in Lymphoma Cell Sensitivity to a Test Drug

This video demonstrates an assay investigating the role of neutrophil-like cells in safeguarding lymphoma cells against the cytotoxicity of chemotherapeutic agents. A 3D co-culture, consisting of lymphoma and neutrophil-like cells, is established and exposed to vincristine—a chemotherapy drug. Direct contact with the neutrophil-like cells induces an upregulation of anti-apoptotic proteins in the lymphoma cells, thereby promoting their survival in the presence of vincristine.
