Lung Pathogens

Lung pathogens are disease-causing microorganisms, including bacteria, viruses, fungi, and parasites, that infect the respiratory tract and can impair breathing and gas exchange. They commonly enter through inhaled droplets or particles, adhere to airway or alveolar surfaces, and trigger tissue injury and inflammation as they multiply or evade host defenses. Studying these pathogens supports accurate diagnosis through microbiological and molecular testing, guides antimicrobial treatment, and informs infection-control and vaccination strategies. Understanding pathogen-specific mechanisms also helps researchers explain pneumonia and other respiratory diseases, improve patient outcomes, and develop therapies for drug-resistant or emerging infections.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.

Lung Capacity

2019

The air in the lungs is measured in volumes and capacities. Lung volume measures reflect the amount of air taken in, released, or left over after a lung function, like a single inhalation. Lung capacity measures are sums of two or more lung volume measures. There are four defined lung volume measures: tidal volume, inspiratory reserve volume, expiratory reserve volume, and residual volume. Tidal volume is the amount of air inhaled and exhaled in a normal breath. Inspiratory...

Development of a Neonatal Piglet Acute Lung Injury Model Recreating the Early Environment of Preterm Infant Lungs

2025

This protocol describes the development of a neonatal piglet model of acute lung injury that models early pathogenic events occurring in the preterm lung, including sub-adequate surfactant quantity, hyperoxia, high-pressure ventilation, and inflammation, to facilitate understanding of molecular triggers of bronchopulmonary dysplasia and enhance therapeutic translation.
