Needle Injection Technique

Needle injection technique is a medical method for delivering medications, vaccines, fluids, or diagnostic substances through a hollow needle into a selected body compartment. After skin preparation and choosing an appropriate needle, the clinician inserts it at a route-specific angle and depth so the material enters the dermal, subcutaneous, muscular, or vascular space; tissue properties and local blood flow then influence distribution and absorption. Used in vaccination, anesthesia, diabetes care, emergency treatment, and diagnostic procedures, the technique depends on accurate site selection, sterile handling, and controlled injection to improve therapeutic effectiveness while reducing pain, tissue injury, infection, and dosing errors.

Direct injection into the rat optic nerve is useful for regenerative research. We demonstrate a minimally-invasive technique for direct injection into a rat optic nerve that does not involve opening the skull. Using this method, surgical complications are minimized and recovery is more rapid.

A Hypertonic Saline Injection Technique for Inducing Glaucoma in Rats

This video demonstrates glaucoma induction in an anesthetized rat by injecting hypertonic saline into the episcleral vein. The saline enters the trabecular meshwork via Schlemm's canal, causing tissue scarring, which blocks aqueous humor outflow and raises intraocular pressure. The elevated pressure damages retinal ganglion cells, leading to glaucoma.

We describe here a protocol for microinjection in the rodent brain that uses quartz needles. These needles do not produce detectable tissue damage and ensure reliable delivery even in deep regions. Moreover, they can be adapted to research needs by personalized designs and can be re-used.
