Retinal Vessel Analysis

Retinal vessel analysis is the quantitative assessment of blood vessels in the retina, providing noninvasive indicators of vascular structure and function that can support medical research and clinical evaluation. The process uses retinal fundus photographs or optical coherence tomography angiography to segment vessels and measure features such as vessel caliber, tortuosity, branching patterns, density, and perfusion, often through automated image-processing algorithms. These measurements can reveal changes associated with diabetic retinopathy, hypertension, cardiovascular disease, and other disorders, helping researchers study disease mechanisms, identify biomarkers, monitor progression, and assess responses to treatment. Standardized analysis may also improve reproducibility across imaging studies and strengthen the role of retinal imaging in precision medicine.

We used retinal samples from retinectomy for a transcriptomic analysis of retinal detachment. We developed a procedure that allows RNA conservation between the surgical blocks and the laboratory. We standardized a protocol to purify RNA by cesium chloride ultracentrifugation to assure that the purified RNAs are suitable for microarray analysis.

We present a protocol for the analysis of coronary vessels in whole embryonic murine hearts up to E15.5, using standard immunological staining methods followed by optical clearance and confocal microscopy. This technique enables visualization of blood vessels throughout the entire heart without the need for time-consuming analysis of serial sections.

The retina shares prominent similarities with the brain and thus represents a unique window to study vasculature and neuronal structure in the brain non-invasively. This protocol describes a method to study dementia using retinal imaging techniques. This method can potentially aid in diagnosis and risk assessment of dementia.
