Three-minute All-out Test

The Three-minute All-out Test is a maximal exercise assessment that measures a person’s ability to produce and sustain power, typically during cycling, while providing insight into both anaerobic and aerobic fitness. During the test, the participant performs a continuous three-minute effort at maximal intensity; rapid phosphagen and glycolytic energy production supports the initial burst, while oxidative phosphorylation increasingly contributes as the effort continues, producing a characteristic decline toward a sustainable power level. Clinicians and exercise researchers use the test to estimate functional capacity, evaluate training responses, and study cardiovascular, metabolic, and neuromuscular performance in health and disease.

We present a protocol to test the aerobic and anaerobic power of the upper body muscles over a duration of 3 min in able-bodied as well as in paraplegic and tetraplegic individuals. The protocol presents specific modifications in its application for upper-body exercise in individuals with or without disability.

We developed novel intrinsic multifunctional nanovesicles called porphysomes, which have structure-dependent fluorescence self-quenching and unique photothermal properties, thus functioning as potent photothermal therapy agents. We formulated porphysomes using high pressure extrusion and investigated their photothermal therapy efficacy in a xenograft tumor model.

EUCAST has developed a direct antimicrobial susceptibility testing (AST) protocol for automated blood cultures. However, its dependence on mass spectrometry-based microbial identification can be obviated by using a direct inoculum preparation protocol in an automated microbial identification system. This approach can provide AST reports within 24 h of sample collection.
