Axon Branching

Axon branching is the formation of new extensions from an axon, enabling a single neuron to connect with multiple target cells and build complex neural circuits. Branches arise when extracellular guidance cues and growth signals regulate the axonal cytoskeleton, prompting actin-rich filopodia to stabilize and recruit microtubules into developing branch sites. In neuroscience, studying axon branching helps explain how neural networks form during development, adapt through experience, and repair after injury. Its regulation is also relevant to understanding neurodevelopmental disorders and to designing strategies that promote precise connectivity and axonal regeneration.

2018

This protocol describes a method for simultaneous imaging of thalamocortical axon branching and synapse formation in organotypic cocultures of the thalamus and cerebral cortex. Individual thalamocortical axons and their presynaptic terminals are visualized by a single cell electroporation technique with DsRed and GFP-tagged synaptophysin.
