Axon Outgrowth

Axon outgrowth is the process by which a neuron extends its axon, enabling the formation of long-range connections essential for nervous system development and repair. At the growing tip, an actin-rich growth cone detects chemical and physical guidance cues, while coordinated rearrangement of actin filaments and microtubules drives membrane extension and forward movement. In neuroscience research, measuring axon outgrowth in cultured neurons helps reveal how extracellular signals, cell adhesion, and intracellular pathways regulate neural connectivity. These studies support investigation of brain development, neurodegenerative disease, spinal cord injury, and strategies to promote axonal regeneration.

Immunofluorescence Assay to Evaluate Effect of Target Proteins on Neurite Outgrowth

In this video, we demonstrate an EGFP-based immunofluorescence assay to identify the efficacy of target proteins on the neurite outgrowth of primary cortical neurons. The primary neurons are transfected with a fluorescent protein, EGFP, and a target protein, and then an immunoassay is performed to investigate the effect of the target protein on neurite outgrowth.

Generation and Culture of Blood Outgrowth Endothelial Cells from Human Peripheral Blood

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.

Dissection and Culture of Chick Statoacoustic Ganglion and Spinal Cord Explants in Collagen Gels for Neurite Outgrowth Assays

We demonstrate how to dissect and culture chick E4 statoacoustic ganglion and E6 spinal cord explants. Explants are cultured under serum-free conditions in 3D collagen gels for 24 hours. Neurite responsiveness is tested with growth factor-supplemented medium and with protein-coated beads.
