Cryo Sectioning

Cryo sectioning is a method for cutting thin slices of frozen biological tissue, enabling researchers to examine anatomy and molecular signals while preserving spatial relationships. In a cryostat, tissue is frozen, embedded in a supportive medium, and sectioned with a sharp blade at low temperature; the resulting slices can be mounted on slides for staining, immunohistochemistry, or imaging. In neuroscience, this approach supports analysis of brain and spinal cord structure, neuronal organization, and the distribution of proteins or labeled cells. By linking cellular features to precise locations, cryo sectioning helps investigate neural circuits, disease-related changes, and responses to experimental treatments.

The present method allows reproducible cryostat sectioning of small, difficult-to-manage, tissue pieces, such as biopsies and brain slices. We utilize a simple aluminum freezing stage to facilitate handling of tissue and a standard cryostat to routinely produce 5-10 micron serial sections from 400 micron thick brain slices.
