Dendritic Protein Labeling

Dendritic protein labeling is a set of methods for marking proteins within neuronal dendrites, enabling researchers to measure their location, abundance, and movement. In fixed cells, fluorescently conjugated antibodies bind target epitopes after fixation and permeabilization, whereas genetically encoded fluorescent tags can report protein distribution and trafficking in living neurons. These approaches reveal how receptors, scaffolding proteins, and cytoskeletal components are organized in dendrites and concentrated at synapses. By linking protein localization to dendritic spine structure and neuronal activity, labeling supports studies of synaptic plasticity, neural development, and mechanisms disrupted in neurological disease.

Dendritic cells uptake antigens and migrate towards immune organs to present processed antigens to T cells. Qdot nanocrystal labeling provides a long-lasting and stable fluorescent signal. This allows tracking of dendritic cells to different organs by fluorescent microscopy.

To understand how complex cell shapes, such as neuronal dendrites, are achieved during development, it is important to be able to accurately assay microtubule organization. Here we describe a robust immunohistological labeling method to examine microtubule organization of dendritic arborization neuron sensory dendrites, trachea, muscle, and other Drosophila larva body wall tissues.

We describe a method to label protein on the surface of living neurons using a specific polyclonal antibody to extracellular epitopes. Protein bound by the antibody on the cell surface and subsequently internalized via endocytosis can be distinguished from protein remaining on, or trafficked to, the surface during the incubation.
