Dorsal Column Injury

Dorsal column injury is damage to the posterior spinal cord pathways that carry fine touch, vibration, and conscious proprioception to the brain, making it important for understanding sensory loss and impaired coordination. The injury disrupts ascending axons from mechanoreceptors; because these fibers cross in the medulla rather than the spinal cord, lesions typically cause reduced vibration and position sense on the same side below the injury. Affected individuals may develop sensory ataxia, unstable gait, and difficulty identifying limb position without visual cues. Studying dorsal column injury supports neurological diagnosis, spinal cord injury research, and development of sensory rehabilitation strategies.

Dorsal Root Ganglion Injection and Dorsal Root Crush Injury as a Model for Sensory Axon Regeneration

This protocol presents the use of a dorsal root ganglion (DRG) injection with a viral vector and a concurrent dorsal root crush injury in an adult rat as a model to study sensory axon regeneration. This model is suitable for investigating the use of gene therapy to promote sensory axon regeneration.

We present a method for chromatin immunoprecipitation from dorsal root ganglia tissue following axonal injury. The approach can be used to identify specific transcription factor binding sites and epigenetic modification of histone and DNA important for the regeneration of injured axons in both the peripheral and central nervous system.

Establishing a Primary Dorsal Root Ganglia Cell Culture from a Rat Spinal Column

This video demonstrates a technique for isolating and culturing dorsal root ganglia (DRG) cells from the rat spinal column. DRGs excised from the spinal column are treated with enzymes and mechanically dissociated to obtain a single-cell suspension. The cells are cultured in a medium containing inhibitors and growth factors that limit glial cell proliferation and promote neuronal growth, respectively.
