Gustatory Response Measurement

Gustatory response measurement is the quantitative assessment of how organisms detect and respond to taste stimuli, providing a window into sensory processing and motivated behavior. In neuroscience, researchers present controlled tastant concentrations to the oral cavity and measure behavioral outputs such as licking, consumption, preference, or rejection, while neural recordings can link these responses to activity in taste pathways. These measurements help distinguish taste sensitivity from appetite and reward, characterize coding of sweet, bitter, salty, sour, and umami qualities, and investigate how learning, disease, drugs, or neural circuit manipulation alter gustatory function.

New Methods to Study Gustatory Coding

We present three new methods to study gustatory coding. Using a simple animal, the moth Manduca sexta (Manduca), we describe a dissection protocol, the use of extracellular tetrodes to record the activity of multiple gustatory receptor neurons, and a system for delivering and monitoring precisely timed pulses of tastants.

In this protocol, we describe two strategies that simultaneously suppress two genes (double gene knockdown) in honey bees. Then we present how to use the proboscis extension response (PER) assay to study the effect of double gene knockdown on honey bee gustatory perception.

The forelegs and proboscis of Drosophila contain a rich repertoire of gustatory sensory neurons. Here, we present a method using calcium imaging to measure physiological responses from sensory neurons in the foreleg and proboscis of live flies upon exogenous application of a gustatory pheromone.
