Hippocampal Implantation

Hippocampal implantation is a neuroscience technique that places electrodes, neural probes, or other experimental devices within the hippocampus to study its role in memory, learning, and neurological disease. During stereotaxic surgery, researchers use anatomical coordinates to guide an implant into a targeted hippocampal region, where it can record neural activity, deliver electrical stimulation, or monitor local circuit responses in living subjects. This approach supports investigations of spatial navigation, memory formation, seizure activity, and hippocampal network function. By linking cellular signals with behavior, hippocampal implantation provides an important foundation for systems neuroscience, disease modeling, and the development of neural interfaces.

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.

We describe the diverse steps to implant chronic silicon probes and to record place cells in mice that are running head-fixed on a cue-enriched treadmill apparatus.

Co-culturing of Ventral Hippocampal and Nucleus Accumbens

This video demonstrates the process of preparing ventral hippocampal and nucleus accumbens tissues from mouse brains for co-culture. Neurons extend projections to form synaptic connections between the ventral hippocampal and nucleus accumbens tissues. This method allows studying neuronal interactions between these two brain regions, offering insights into cognition, behavior, and neurological disorders.
