Neuronal Structure Imaging

Neuronal structure imaging comprises microscopy methods used to visualize the morphology and organization of neurons, making features such as cell bodies, dendrites, axons, and synaptic connections accessible for study. These methods typically use fluorescent labels or genetically encoded markers combined with light or electron microscopy to capture cellular structure, often through optical sectioning and three-dimensional reconstruction. In neuroscience, neuronal structure imaging helps researchers examine brain development, circuit organization, synaptic remodeling, and structural changes associated with injury or disease. Quantitative analysis of neuronal shape and connectivity also supports investigations of how anatomy relates to neural function and behavior.

Imaging Dendritic Spines of Rat Primary Hippocampal Neurons using Structured Illumination Microscopy

This article describes a working protocol to image dendritic spines from hippocampal neurons in vitro using Structured Illumination Microscopy (SIM). Super-resolution microscopy using SIM provides image resolution significantly beyond the light diffraction limit in all three spatial dimensions, allowing the imaging of individual dendritic spines with improved detail.

Imaging the dynamic behavior of organelles and other subcellular structures in vivo can shed light on their function in physiological and disease conditions. Here, we present methods for genetically tagging two organelles, centrosomes and mitochondria, and imaging their dynamics in living zebrafish embryos using wide-field and confocal microscopy.
