Optic Nerve Stump

The optic nerve stump is the remaining segment of the optic nerve after it has been severed, injured, or surgically transected, providing a model for studying visual pathway damage and repair. Following injury, retinal ganglion cell axons undergo degeneration, while inflammatory and glial responses alter the stump’s cellular environment and can influence axonal regeneration. In neuroscience research, optic nerve stump models help investigators examine neuronal survival, Wallerian degeneration, glial scar formation, and strategies for restoring connections between the retina and brain. Findings from these studies support the development of experimental treatments for traumatic optic neuropathy and other conditions involving optic nerve damage.

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.

Optic Nerve transection is a widely used model of adult CNS injury. Ninety percent of retinal ganglion cells (RGCs) whose axons are completely transected (axotomy) die within 14 days after axotomy. This model is easily amenable to experimental manipulations and highly reproducible.

Direct injection into the rat optic nerve is useful for regenerative research. We demonstrate a minimally-invasive technique for direct injection into a rat optic nerve that does not involve opening the skull. Using this method, surgical complications are minimized and recovery is more rapid.
