Presynaptic Ion Currents

Presynaptic ion currents are the time-dependent movements of charged ions across a neuron’s presynaptic membrane, where they regulate synaptic signaling and neurotransmitter release. When an action potential depolarizes the axon terminal, voltage-gated calcium channels open, and calcium influx through its electrochemical gradient triggers synaptic-vesicle fusion; sodium and potassium currents help shape terminal excitability and timing. Researchers measure these currents with patch-clamp and voltage-clamp electrophysiology to characterize ion-channel function, release probability, and short-term synaptic plasticity. Their study clarifies how neurons communicate and supports research into circuit function, neurological disorders, and drugs that alter synaptic transmission.

Presynaptically Silent Synapses Studied with Light Microscopy

Glutamatergic synapses can switch from an active mode to a silent mode. We demonstrate that presynaptic activity status in dissociated culture of rodent neurons is visualized using a fixable form of the FM1-43 dye to visualize active synapses and immunostaining with vGluT-1 antibody to visualize all glutamate synapses.

GABAergic presynaptic inhibition is a powerful inhibitory mechanism in the spinal cord important for motor and sensory signal integration in spinal cord networks. Underlying primary afferent depolarization can be measured by recording of dorsal root potentials (DRP). Here we demonstrate a method of in vivo recording of DRP in mice.

Vibrodissociation of Neurons from Rodent Brain Slices to Study Synaptic Transmission and Image Presynaptic Terminals

This report demonstrates a technique for mechanical isolation of individual viable neurons retaining attached presynaptic boutons. Vibrodissociated neurons have the advantages of rapid production, excellent pharmacological control and improved space-clamp without influence from neighboring cells. This method can be used for imaging of synaptic elements and patch-clamp recording.
