Quantitative Assay

A quantitative assay is a measurement method that determines the amount or concentration of a target substance, particle, or measurable component in a sample, providing numerical results rather than a qualitative presence-or-absence judgment. It works by converting a sample response, such as intensity, absorbance, fluorescence, mass, or electrical signal, into a quantity through comparison with reference standards and a calibration curve under controlled conditions. In physics, quantitative assays support precise measurements of radiation, materials, particles, and instrument responses, while uncertainty analysis and calibration help assess reliability, compare experiments, and connect observed signals with underlying physical properties.

Optimization of a Quantitative Micro-neutralization Assay

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

Here, we present protocols to rapidly screen and characterize enzymes for antimicrobial activity. The microslide diffusion assay and the dye-release assay utilize target bacterial substrates for qualitative and quantitative enzymatic activity evaluation.

The phagokinetic motility track assay is a method used to assess the movement of cells. Specifically, the assay measures chemokinesis (random cell motility) over time in a quantitative manner. The assay takes advantage of the ability of cells to create a measurable track of their movement on colloidal gold-coated coverslips.

A Quantitative Assay for Insulin-expressing Colony-forming Progenitors

A three-dimensional clonogenic assay that allows pancreatic-like progenitors to differentiate into insulin-expressing colonies is described. This method takes advantage of semi-solid media containing methylcellulose, Matrigel and growth factors, in which single progenitors proliferate and differentiate in vitro, permitting quantification of the number of functional progenitors in a population.
