W komórkach prokariotycznych operon to grupa sekwencji genetycznych, które zawierają elementy regulatorowe i kilka genów kodujących białka, genów strukturalnych, które są transkrybowane razem. Zwykle, gdy stężenie glukozy jest obfite, białko represorowe lac ściśle wiąże się z operatorem i zapobiega transkrypcji genów lac poprzez blokowanie wiązania polimerazy RNA z promotorem. Represor lac ulega konstytutywnej ekspresji, co oznacza, że kodujący go gen jest domyślnie włączony. Gdy stężenie glukozy jest bardzo niskie, komórka będzie wykorzystywać laktozę jako źródło energii. Dodatkowo, przy niskim poziomie glukozy, zwiększa się ilość cyklicznego AMP cząsteczki sygnałowej i wiąże się z białkiem aktywatora katabolitu lub CAP. Razem wiążą się z sekwencją regulatorową, proksymalnie przed promotorem i pomagają rekrutować polimerazę RNA w celu znacznego zwiększenia transkrypcji. Operons

Prokarioty mogą kontrolować ekspresję genów poprzez operony — sekwencje DNA składające się z elementów regulatorowych i skupionych, funkcjonalnie powiązanych genów kodujących białka. Operony wykorzystują pojedynczą sekwencję promotora do inicjowania transkrypcji klastra genów (tj. grupy genów strukturalnych) do pojedynczej cząsteczki mRNA. Sekwencja terminatora kończy transkrypcję. Sekwencja operatora, zlokalizowana pomiędzy promotorem a genami strukturalnymi, uniemożliwia aktywność transkrypcyjną operonu, jeśli jest on związany przez białko represorowe. W sumie promotor, operator, geny strukturalne i terminator tworzą rdzeń operonu.

Operony są zwykle indukowalne lub tłumione. Operony indukowalne, takie jak operon bakteryjny lac, są zwykle “wyłączone” ale “włączają się” w obecności małej cząsteczki zwanej induktorem (np. allolaktozą). Gdy nie ma glukozy, ale obecna jest laktoza, allolaktoza wiąże się i inaktywuje represor—operonu lac, umożliwiając operonowi wytwarzanie enzymów odpowiedzialnych za metabolizm laktozy.

Operony podlegające represji, takie jak operon bakteryjny trp, są zwykle “włączone”, ale “wyłączają” się w obecności małej cząsteczki zwanej korepresorem (np. tryptofanu). Kiedy tryptofan — niezbędny aminokwas — jest obfity, tryptofan wiąże się i aktywuje represor— trp, uniemożliwiając operonowi wytwarzanie enzymów niezbędnych do jego syntezy.

Operony mogą być również konstytutywne (tj. ciągłe) aktywne. Na przykład operony bakteryjnego rybosomalnego RNA (rRNA) są zawsze “włączone” ponieważ rRNA są stale potrzebne do translacji.

Inne elementy regulacyjne również przyczyniają się do skoordynowanej ekspresji genów operonu. Geny regulatorowe kodują białka aktywujące transkrypcję lub represory. Na przykład geny lacI i trpR kodują odpowiednie represory operonu. Dodatkowe sekwencje regulatorowe, takie jak miejsce wiązania białka aktywatora katabolizmu operonu lac (CAP), zapewniają miejsca wiązania dla innych aktywatorów lub represorów.
