Membrane Protein Diffusion

Membrane protein diffusion is the movement of proteins within or across biological membranes, a process that shapes cellular signaling, transport, adhesion, and organization. In biochemistry, most diffusion occurs laterally through the lipid bilayer, driven by thermal motion and influenced by membrane fluidity, protein size, lipid interactions, and attachment to the cytoskeleton or extracellular structures. Researchers measure these dynamics using approaches such as fluorescence recovery after photobleaching and single-molecule tracking, which reveal mobility, confinement, and binding behavior. Understanding membrane protein diffusion helps explain how cells coordinate receptors and channels and supports research on signaling defects, membrane organization, and therapeutic targeting.

Protein Diffusion in the Membrane

2023

Proteins show rotational as well as lateral diffusion across the membrane. The lateral diffusion of proteins was confirmed through the cell fusion experiment where mouse and human cells were fused, resulting in hybrid cells. When the human and mouse cells fused, the specific membrane proteins on human and mouse cells were marked with the red and green-fluorescent markers, respectively. Initially, the red and green fluorescence was located on the respective hemisphere of the cell. As time...

Reconstitution of Membrane Proteins

2023

Reconstitution is the process of returning an isolated biomolecule to its original form or function. This is particularly useful for studying membrane proteins, which enable important cellular functions and affect the behavior of nearby lipids. To study the function of purified membrane proteins in situ, they must be reconstituted by integrating them into an artificial lipid membrane. This video introduces membrane protein reconstitution concepts and related procedures, such as protein...
