Quantitative Cell Analysis

Quantitative cell analysis is the measurement and statistical evaluation of cellular features, activities, and responses to convert biological observations into numerical data. It combines methods such as microscopy, flow cytometry, image analysis, and biochemical assays to quantify parameters including cell number, size, fluorescence, protein abundance, viability, and metabolic activity under defined conditions. In biochemistry, these measurements reveal how molecular pathways regulate cell behavior and how cells respond to nutrients, drugs, genetic changes, or environmental stress. Quantitative analysis supports reproducible experiments, enables comparisons across samples, and helps connect molecular mechanisms with phenotypes in research, diagnostics, and therapeutic development.

A Quantitative Fitness Analysis Workflow

Quantitative Fitness Analysis (QFA) is a complementary series of experimental and computational methods for estimating microbial culture fitnesses. QFA estimates the effect of genetic mutations, drugs or other applied treatments on microbe growth. Experiments scaling from focussed analysis of single cultures to thousands of parallel cultures can be designed.

In this protocol, we present the experimental procedures of a cell spreading assay that is based on live-cell microscopy. We provide an open-source computational tool for the unbiased segmentation of fluorescently labeled cells and quantitative analysis of lamellipodia dynamics during cell spreading.

Improved Visualization and Quantitative Analysis of Drug Effects Using Micropatterned Cells

Adhesive micropatterns that normalize cellular architecture can be used to increase sensitivity in the detection of drug effects, improve reproducibility and simplify automated image acquisition and analysis. Such technology will benefit drug/siRNA screening assays, performed on conventional cell culture supports and consequently suffering from excessive cell-to-cell variability.

Live-cell Imaging and Quantitative Analysis of Embryonic Epithelial Cells in Xenopus laevis

Xenopus embryonic epithelia are an ideal model system to study cell behaviors such as polarity development and shape change during epithelial morphogenesis. Traditional histology of fixed samples is increasingly being complemented by live-cell confocal imaging. Here we demonstrate methods to isolate frog tissues and visualize live epithelial cells and their cytoskeleton using live-cell confocal microscopy.
