Amphiphilic Peptoids

Amphiphilic peptoids are synthetic, peptide-like oligomers that combine water-compatible and water-avoiding side chains, making them useful materials for bioengineering. Built on an N-substituted glycine backbone, they can fold and self-assemble in aqueous environments as hydrophobic segments cluster away from water while hydrophilic groups remain exposed. This tunable organization produces structures such as micelles, membrane-like assemblies, and nanofibers whose size, stability, and surface properties can be adjusted through sequence design. Amphiphilic peptoids therefore support research into drug delivery, biomimetic membranes, antimicrobial materials, and other engineered systems that require control over molecular recognition and interactions with biological environments.

A protocol to synthesize peptoids with mixed cationic functionality in the same sequence is presented (lysine- and arginine-type monomers). Subsequent testing of these compounds against Leishmania mexicana, the protozoan parasites that cause cutaneous leishmaniasis, is also described.

A simple and general manual peptoid synthesis method involving basic equipment and commercially available reagents is outlined, enabling peptoids to be easily synthesized in most laboratories. The synthesis, purification and characterization of an amphiphilic peptoid 36mer is described, as well as its self-assembly into highly-ordered nanosheets.

The key steps of living anionic polymerization of phenyl glycidyl ether (PheGE) on methoxy-polyethylene glycol (mPEG-b-PPheGE) are described. The resulting block copolymer micelles (BCMs) were loaded with doxorubicin 14% (wt%) and sustained release of drug over 4 days under physiologically relevant conditions was obtained.
