Digital Reconstruction Algorithms

Digital reconstruction algorithms are computational methods that convert incomplete, indirect, or two-dimensional measurements into representations of three-dimensional biological structures, processes, or tissues. They typically combine image processing, segmentation, geometric modeling, and optimization to align data, infer missing features, and generate a coherent reconstruction from sources such as microscopy, medical imaging, or sensor measurements. In bioengineering, these algorithms support anatomical modeling, tissue and organoid analysis, biomaterial design, and simulation-based research. Their accuracy influences how researchers quantify structure, plan interventions, evaluate engineered tissues, and connect experimental observations with predictive computational models.

Reconstruction of 3-Dimensional Histology Volume and its Application to Study Mouse Mammary Glands

We present an image registration approach for 3-dimensional (3D) histology volume reconstruction, which facilitates the study of the changes of an organ at the level of macrostructures made up of cells . Using this approach, we studied the 3D changes between wild-type and Igfbp7-null mammary glands.

Compact Lens-less Digital Holographic Microscope for MEMS Inspection and Characterization

We present a compact reflection digital holographic system (CDHM) for inspection and characterization of MEMS devices. A lens-less design using a diverging input wave providing natural geometrical magnification is demonstrated. Both static and dynamic studies are presented.

A workflow for creating three-dimensional (3D) virtual hybrid models has been designed based on cone-beam computed tomography dataset and intraoral optical scans utilizing radiographic image segmentation methods and free-form surface modeling. Digital models are used for the virtual planning of reconstructive dentoalveolar surgical procedures.

Rodent skilled reaching is commonly used to study dexterous skills, but requires significant time and effort to implement the task and analyze the behavior. We describe an automated version of skilled reaching with motion tracking and three-dimensional reconstruction of reach trajectories.
