Biofilm Quantification

Biofilm quantification is the measurement of the biomass, cell number, or activity of microorganisms growing within a surface-associated community, providing essential data for bioengineering and microbial research. Methods such as crystal violet staining estimate total attached biomass, while viable-cell counting, metabolic assays, optical density measurements, and microscopy assess different aspects of biofilm development. Because each method captures a distinct feature, researchers often combine approaches to distinguish living cells from extracellular polymeric substances and inactive material. These measurements support the evaluation of biomaterials, antimicrobial treatments, bioreactors, wastewater systems, and engineered microbial communities, enabling comparisons of adhesion, maturation, and treatment effectiveness.

Microtiter Dish Biofilm Formation Assay

The assay describes a rapid means to measure early biofilm formation in bacteria and fungi. This method uses a microtiter plate as the substratum for microbial biofilm formation, and the biofilm is visualized using crystal violet strain. The assay provides either a qualitative or quantitative assay for early biofilm formation.

Mycobacterium tuberculosis forms drug tolerant biofilms when cultured in certain conditions. Here we describe methods for culturing M. tuberculosis biofilms and determining the frequency of drug tolerant persisters. These protocols will be useful for further studies into the mechanisms of drug tolerance in M. tuberculosis.
