Human Renal Cells

Human renal cells are specialized cells of the kidney that maintain fluid, electrolyte, and waste balance through filtration, transport, secretion, and endocrine signaling. In structures such as nephrons, glomerular cells filter blood while tubular epithelial cells selectively reabsorb essential substances and secrete unwanted compounds, processes shaped by cell polarity, membrane transporters, and interactions with the extracellular matrix. In bioengineering, these cells support kidney-on-a-chip systems, engineered tissues, organoids, and disease models that reproduce renal physiology under controlled conditions. Such platforms help evaluate nephrotoxicity, study kidney disorders, and guide the development of regenerative therapies and implantable renal devices.

Development of Human Renal Tubular Epithelial Cell Primary Cultures in Monolayers and Three-Dimensional Conditions

2025

Here, we describe the protocols for obtaining primary cultures of human renal tubular epithelial cells (HRTEC) from the kidney cortex to develop monolayers and three-dimensional cultures on an extracellular matrix (3D-HRTEC).

A protocol is reported here for the selective ablation of renal macrophages to study their regeneration using the human CD59/intermedilysin cell ablation tool. This method is also applicable for studying the function and regeneration of the other cell populations in the kidney, liver, and fatty tissue.
