Lateral Flow Assay

Lateral flow assays are portable paper-based diagnostic tests that detect specific biological targets rapidly and with minimal equipment. A liquid sample moves by capillary action through a porous strip, where labeled binding molecules interact with the target and form visible lines at immobilized capture zones; control lines verify proper flow and test function. In bioengineering, lateral flow assays support point-of-care detection of proteins, nucleic acids, pathogens, and other biomarkers. Their low cost, simple operation, and rapid visual readout make them valuable for decentralized testing, while ongoing innovations in materials, signal amplification, and quantitative readers aim to improve sensitivity and expand their research and clinical applications.

An Assay for Lateral Line Regeneration in Adult Zebrafish

Because many zebrafish models of neurological and non-neurological diseases are studied in the adult fish rather than the embryo/larvae, we developed a quantitative lateral line regenerative assay that can be applied to adult zebrafish disease models. The assay involved resolution at the 1) neuromast and 2) individual hair cell levels.

Detection of Invasive Pulmonary Aspergillosis in Haematological Malignancy Patients by using Lateral-flow Technology

A rapid and accurate point-of-care test for invasive pulmonary aspergillosis is presented. It takes advantage of lateral-flow technology using a specific monoclonal antibody that binds to an Aspergillus antigen secreted during pulmonary infections. The assay is compatible with serum and brochoalveolar lavage and represents a novel adjunct test for disease diagnosis.

The micronucleus (MN) assay is a well-established test for quantifying DNA damage. However, scoring the assay using conventional techniques such as manual microscopy or feature-based image analysis is laborious and challenging. This paper describes the methodology to develop an artificial intelligence model to score the MN assay using imaging flow cytometry data.

Fluorescent Lateral Flow Immunoassay Based on Quantum Dots Nanobeads

Here, we describe a protocol for the preparation of quantum dot nanobeads (QDNB) and the detection of disease biomarkers using QDNB-based lateral flow immunoassay strips. The test results can be qualitatively assessed under UV light illumination and quantitatively measured using a fluorescent strip reader within 15 min.

Human Neutrophil Flow Chamber Adhesion Assay

A method of quantitating neutrophil adhesion is reported. This method creates a dynamic flow environment similar to that encountered in a blood vessel. It allows the investigation of neutrophil adhesion to either purified adhesion molecules (ligand) or endothelial cell substrate (HUVEC) in a context similar to the in vivo environment with sheer stress.
