Enzymatic Activity Assay

An enzymatic activity assay is a laboratory method that measures how efficiently an enzyme converts a substrate into one or more products, providing a functional view of enzyme performance. The assay typically monitors substrate depletion or product formation under controlled conditions, such as defined substrate concentration, pH, temperature, and reaction time, often through a measurable color, fluorescence, or absorbance change. Researchers use these assays to compare enzyme activity, characterize reaction kinetics, evaluate inhibitors or activators, and assess how mutations or experimental conditions affect function. As a core biological technique, enzymatic activity assays support studies of metabolism, molecular biology, drug development, and disease mechanisms.

This video demonstrates the dye release assay for quantifying the enzymatic activity of antimicrobial proteins. The antimicrobial protein degrades the bacteria's dye-labeled cell wall and releases the dye molecules turning the solution blue. The absorbance of the solution correlates with the enzymatic activity of the antimicrobial protein.

G4 Resolvase1 binds to G-quadruplex (G4) structures with the tightest reported affinity for a G4-binding protein and represents the majority of the G4-DNA unwinding activity in HeLa cells. We describe a novel protocol that harnesses the affinity and ATP-dependent unwinding activity of G4-Resolvase1 to specifically purify catalytically active recombinant G4R1.
