Agar Plate Spraying Technique

The agar plate spraying technique is a microbiological method for distributing a liquid sample across the surface of solidified agar, enabling consistent growth and analysis of microorganisms. A bacterial, fungal, or other biological suspension is atomized through a spray device and applied evenly to the plate, where cells remain on the nutrient surface and form colonies during incubation. This approach can improve coverage and reproducibility compared with localized inoculation, supporting colony isolation, microbial screening, viability assessment, and studies of growth responses. Careful control of suspension concentration, spray pattern, and incubation conditions helps produce interpretable results.

When working with media and reagents used to culture microorganisms, aseptic technique must be practiced to ensure contamination is minimized. A variety of plating methods are routinely used to isolate, propagate, or enumerate bacteria and phage, all of which incorporate procedures that maintain the sterility of experimental materials.

Topical Airway Anesthesia for Awake-endoscopic Intubation Using the Spray-as-you-go Technique with High Oxygen Flow

The topical anesthetic lidocaine was atomized using a high oxygen flow through the working channel of a flexible intubating endoscope to achieve topical airway anesthesia for awake endotracheal intubation. We prefer this modified spray-as-you-go technique for endoscopic intubation to classical bolus application because of higher patient satisfaction and better compliance.
