Bacterial Pellet

A bacterial pellet is the concentrated mass of bacterial cells collected from a liquid culture, typically after centrifugation, and is a common starting material in biology laboratories. During centrifugation, centrifugal force drives cells and other dense particles toward the bottom of a tube, where they compact into a visible pellet while the surrounding liquid forms the supernatant. Researchers can remove the supernatant and resuspend the pellet for cell washing, microscopy, protein extraction, DNA or RNA isolation, or downstream biochemical analysis. Pellet size and appearance can provide a useful, though approximate, indication of cell recovery and culture density.

Liquid-grown Streptomyces cultures are characterized by mycelial pellets that are heterogeneous in size. We here describe a method to analyze and sort such pellets in a high-throughput manner. These pellets can be used for further analyses, which will provide leads to understand and control growth heterogeneity.

2016

In this study, a protocol was developed to produce good quality pellets using a flat die pellet mill at reduced specific energy consumption testing high-moisture corn stover and a starch based binder. The results indicated that adding a corn starch binder improved the pellet durability, reduced percent fines and decreased specific energy consumption.

Microplastics act as vector of potentially toxic organic contaminants with unpredictable effects. This protocol describes an alternative methodology for assessing the levels of organochlorine pesticides adsorbed on plastic pellets and identifying the polymer chemical structure. The focus is on pressurized fluid extraction and attenuated total reflectance Fourier transform infrared spectroscopy.
