Carbon-14 Palmitic Acid

Carbon-14 palmitic acid is a radiolabeled form of palmitic acid, a saturated 16-carbon fatty acid, used to trace lipid metabolism in biological systems. Cells incorporate the molecule into lipid stores or activate it to palmitoyl-CoA, which can undergo mitochondrial beta-oxidation, releasing labeled carbon through successive cleavage of two-carbon acetyl-CoA units. Researchers use the radioactive signal to measure fatty acid uptake, incorporation, oxidation, and energy production in cultured cells, tissues, and organisms. These measurements help clarify metabolic pathways and reveal how nutrition, hormones, disease, or experimental treatments alter lipid utilization.

This paper describes a method for the measurement of fuel oxidation in Drosophila melanogaster in which trace amounts of specific radiolabeled metabolic substrates are fed to flies. The exhaled radiolabeled CO2 that is a produced from fuel oxidation is collected and measured.

We present the benzene polycarboxylic acid (BPCA) method for assessing pyrogenic carbon (PyC) in the environment. The compound-specific approach uniquely provides simultaneous information about the characteristics, quantity and isotopic composition (13C and 14C) of PyC.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.
