Competitive Inhibitor Screen

A competitive inhibitor screen is an enzymatic assay used to identify and characterize molecules that reduce activity by competing with a substrate for an enzyme’s active site. The assay measures reaction rates across defined substrate and inhibitor concentrations, then compares the resulting kinetics with an uninhibited control; increasing substrate can lessen inhibition, while competitive binding typically increases the apparent Km without changing Vmax. In biology and drug discovery, these screens help determine inhibitor potency, estimate kinetic parameters, assess substrate-site interactions, and prioritize compounds for further study. The results can clarify enzyme function and guide development of selective therapeutics.
