Matrix Invasion Assay

A matrix invasion assay is an in vitro method for measuring how effectively cells move through an extracellular matrix barrier, providing a controlled model of tissue invasion. In a typical setup, cells are placed on a matrix-coated porous membrane, while a chemoattractant in the lower chamber promotes directional migration; invasive cells cross the matrix and membrane, where they can be stained and quantified. The assay is widely used to study cell motility, extracellular matrix remodeling, and metastatic potential in cancer biology. It also supports evaluation of signaling pathways and candidate compounds that alter invasive behavior.

Tumor microenvironment is an essential part of cancer growth and invasion. To mimic carcinoma progression, a biologically relevant human matrix is needed. This protocol introduces an improvement for the in vitro three-dimensional spheroid invasion assay by applying a human leiomyoma-based matrix. The protocol also introduces a computer-based cell invasion analysis.
