Nanodisc Reconstitution

Nanodisc reconstitution is a technique for embedding membrane proteins within small, soluble lipid bilayers that mimic the native cell membrane. It works by combining purified membrane proteins with phospholipids and scaffold molecules, then adjusting their proportions and assembly conditions so the lipids form a stable disc around the protein. This controlled environment preserves important structural and functional features while improving access for biochemical and biophysical measurements. In biology, nanodisc reconstitution supports studies of membrane-protein folding, transport, signaling, and interactions with drugs or other biomolecules, providing a useful platform for investigating proteins that are difficult to analyze in detergent solutions.

Reconstitution of Membrane Proteins

2023

Reconstitution is the process of returning an isolated biomolecule to its original form or function. This is particularly useful for studying membrane proteins, which enable important cellular functions and affect the behavior of nearby lipids. To study the function of purified membrane proteins in situ, they must be reconstituted by integrating them into an artificial lipid membrane. This video introduces membrane protein reconstitution concepts and related procedures, such as protein...

This study outlines the procedure for reconstituting the membrane protein TWIK-related acid-sensitive K+ channel 2 (Task2) into nanodiscs. The successful assembly was confirmed by single-particle cryo-electron microscopy, which yielded well-defined two-dimensional class averages.

Efforts to isolate the catalytic subunit of telomerase, TERT, in sufficient quantities for structural studies, have been met with limited success for more than a decade. Here, we present methods for the isolation of the recombinant Tribolium castaneum TERT (TcTERT) and the reconstitution of the active T. castaneum telomerase ribonucleoprotein (RNP) complex in vitro.
