Osteocyte Markers

Osteocyte markers are genes, proteins, and cellular features used to identify osteocytes, the mature bone cells embedded within the mineralized matrix and central regulators of bone remodeling. These markers reflect osteocyte differentiation and function, including expression of SOST, DMP1, PHEX, FGF23, and E11/gp38, as well as development of dendritic processes within the lacunocanalicular network. In biology research, marker panels help distinguish osteocytes from osteoblasts and other bone cells in tissue sections, cultured models, and single-cell analyses. They support studies of skeletal development, mineral metabolism, mechanical sensing, and diseases in which osteocyte signaling alters bone formation or resorption.

Treatment of primary mouse osteoblasts with retinoic acid produces a homogeneous population of ramified cells bearing morphological and molecular features of osteocytes. The method overcomes the difficulty of obtaining and maintaining primary osteocytes in culture, and can be advantageous to study cells derived from transgenic...

This study outlines the method to visualize and develop three-dimensional (3D) models of osteocytes within the lacunar-canalicular network (LCN) for computational fluid dynamics (CFD) analysis. The generated models using this method help to understand osteocyte mechanosensation in healthy or diseased bones.

Multiplexed Immunofluorescence Assay for Spatial Assessment of Senescence Markers in vivo

2026

Multiplexed immunofluorescence enables sequential labeling of multiple antibody markers within a single tissue section to support spatial analysis of tissue architecture and cell populations. Here, a workflow for multiplex detection of senescence-associated, structural, and immune markers is presented, including manual and automated slide preparation, iterative imaging, and single-cell analysis using digital pathology software.

Here, we present protocols for analyzing bone remodeling within a lab-on-a-chip platform. A 3D printed mechanical loading device can be paired with the platform to induce osteocyte mechanostransduction by deforming the cellular matrix. The platform can also be used to quantify bone remodeling functional outcomes from osteoclasts and osteoblasts (resorption/formation).
