In Utero Culture

In utero culture is a specialized biological approach that maintains developing embryos, fetuses, or fetal tissues within the uterus to study growth under physiologically relevant conditions. The uterine environment supports development through maternal nutrients, oxygen exchange, hormonal signals, and tissue interactions mediated largely by the placenta, while researchers may introduce controlled experimental treatments or genetic manipulations. This approach supports research in embryology, developmental biology, reproductive science, and prenatal toxicology by preserving key features of gestation that are difficult to reproduce in laboratory culture systems. It can also help assess developmental outcomes and inform future studies of prenatal therapies.

Here we present a protocol providing a tool to examine regulatory mechanisms of specific genes during hippocampal development. Employing ex utero electroporation and organotypic slice culture allows the up- and down-regulation of the expression of genes of interest in single cells and follow their fate during development.
