Paracrine Ros

Paracrine ROS refers to reactive oxygen species that act as signals between neighboring cells, extending redox communication beyond the cell in which they are produced. In this process, molecules such as hydrogen peroxide diffuse through the extracellular space or cross cell membranes through aquaporin channels, where they reversibly modify redox-sensitive proteins and alter signaling pathways in recipient cells. Paracrine ROS helps coordinate processes including inflammation, tissue repair, vascular responses, and cellular stress adaptation. Studying this form of intercellular communication clarifies how localized oxidative events influence surrounding tissues and may reveal mechanisms involved in disease progression and therapeutic response.

2011

An efficient method to gain insights into visualizing the paracrine-derived ROS induction of endothelial Ca2+ signaling is described. This method takes advantage of measuring paracrine derived ROS triggered Ca2+ mobilization in vascular endothelial cells in a co-culture model.

ROS Detection with DCFH-DA Staining: Measuring Total ROS in Colorectal Cancer Cell Lines

2023

This video describes the protocol for detecting total cellular reactive oxygen species (ROS) using 2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA). This method helps visualize ROS localization in the adherent cells using a fluorescence microscope and further quantify ROS intensity with a fluorescence plate reader.

Paracrine Signaling

2021

Paracrine signaling allows cells to communicate with their immediate neighbors via secretion of signaling molecules. Such a signal can only trigger a response in nearby target cells because the signal molecules degrade quickly or are inactivated if not taken up. Prominent examples of paracrine signaling include nitric oxide signaling in blood vessels, synaptic signaling of neurons, the blood clotting system, tissue repair/wound healing, and local allergic skin reactions.Nitric oxide as a...
