Activated Fibroblasts

Activated fibroblasts are connective-tissue cells that shift from a resting, tissue-maintenance state to a matrix-producing and signaling state in response to injury, inflammation, or disease. In cancer, tumor-derived factors such as transforming growth factor beta stimulate fibroblast activation, leading to contractile features, extracellular-matrix deposition, tissue remodeling, and secretion of cytokines and growth factors. These cancer-associated fibroblasts help shape the tumor microenvironment by influencing cancer-cell growth, invasion, blood-vessel formation, and immune-cell behavior. Studying activated fibroblasts clarifies how stromal cells support tumor progression and may guide strategies that alter the extracellular matrix or disrupt fibroblast-mediated therapy resistance.

Cancer Associated Fibroblasts (CAFs) facilitate tumor initiation, growth and progression through signaling that promotes proliferation, angiogenesis, and inflammation. Here we describe a method to isolate pure populations of normal fibroblasts and CAFs from fresh mouse and human tissues by cell sorting, using PDGFRα as a surface marker.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.
