Cfse-labeled Tumor Complex

A CFSE-labeled tumor complex is a tumor-cell preparation marked with carboxyfluorescein succinimidyl ester (CFSE), a fluorescent tracer that enables researchers to follow tumor cells in cancer research. CFSE enters cells and forms stable covalent bonds with intracellular amines, while its fluorescence is approximately divided between daughter cells during cell division. Fluorescence-based analysis can therefore reveal tumor-cell proliferation, persistence, and distribution within complex experimental systems. This approach supports studies of tumor growth, cell-cell interactions, and responses to anticancer treatments by linking detectable fluorescence patterns with changes in tumor-cell behavior.

CFSE covalently labels long-lived intracellular molecules with the fluorescent dye, carboxyfluorescein. As such, when a CFSE-labeled cell divides, its progeny have half the amount of fluorescence, which can thereby be used to assess cell division. This article describes the procedures typically used for labeling mouse lymphocytes with CFSE.
