Microvasculature Assessment

Microvasculature assessment is the evaluation of small blood vessels and their structure, density, blood flow, or permeability, providing insight into tissue health and disease. In cancer research, it commonly uses imaging or quantitative measurements to characterize tumor-associated vessels, where angiogenic signaling can promote irregular vessel formation and altered perfusion. These assessments help researchers examine how the tumor microenvironment supports growth, invasion, and treatment resistance, while enabling comparison of vascular changes during disease progression or therapy. Measuring microvascular features can therefore support studies of tumor biology, biomarker development, drug delivery, and responses to antiangiogenic or other cancer treatments.

This video describes the technique of DCE-MRI for orthotropic pancreatic tumor xenografts in mice. DCE-MRI is a non-invasive method to analyze microvasculature in a target tissue, and useful to assess vascular response in a tumor following a novel therapy. This method will assist investigators to apply DCE-MRI for orthotopic gastrointestinal cancer mouse models.

Spatial Temporal Analysis of Fieldwise Flow in Microvasculature

To quantify microvascular flow from high speed capillary flow image sequences, we developed STAFF (Spatial Temporal Analysis of Fieldwise Flow) software. Across the full image field and over time, STAFF evaluates flow velocities and generates a sequence of color-coded spatial maps for visualization and tabular output for quantitative analyses.

Humanized Mouse Model to Study Bacterial Infections Targeting the Microvasculature

Neisseria meningitidis is a human specific pathogen that infects blood vessels. In this protocol human microvessels are introduced into a mouse by grafting human skin onto immunocompromised mice. Bacteria adhere extensively to the human vessels, leading to vascular damage and development of the purpuric rash typically observed in human cases.

Ultrasound Localization Microscopy (ULM) is a recently developed technique that allows for an unprecedented level of resolution in imaging the microvasculature in vivo. In this work, we describe in detail how to obtain super-resolved images of the brain microvasculature in rodents using a standardized functional ultrasound imaging platform designed for preclinical research.
