Ion Mobility

Ion mobility is a gas-phase separation technique that distinguishes ions according to how they move through a buffer gas under the influence of an electric field. An ion’s drift depends on its charge, size, shape, and interactions with the surrounding gas, allowing species with similar mass-to-charge ratios to be separated by their mobility. In chemistry, ion mobility is often coupled with mass spectrometry to add structural and compositional information, including collision cross section measurements. These capabilities support the analysis of complex mixtures, molecular conformations, isomers, biomolecules, and reaction products, strengthening identification and characterization in analytical research.

Ion mobility spectrometry (IMS) is an interesting complement to mass spectrometry for the characterization of biomolecules, notably because it is sensitive to isomerism. This protocol describes a tandem IMS (IMS/IMS) experiment, which allows the isolation of a molecule and the generation of the mobility profiles of its fragments.

Ion mobility-mass spectrometry is an emerging gas-phase technology that separates ions, based on their collision cross-section and mass. The method provides three-dimensional information on the overall topology and shape of protein complexes. Here, we outline a basic procedure for instrument setting and optimization, calibration of drift times, and data interpretation.
