Talar Tilt Test

The Talar Tilt Test is a clinical examination used to assess ankle ligament stability, particularly after a sprain or other injury. With the patient’s leg stabilized, the examiner tilts the talus within the ankle mortise by moving the foot into inversion or eversion and compares laxity and pain with the uninjured side. Excessive inversion may indicate injury to the lateral ankle ligaments, especially the calcaneofibular ligament, whereas abnormal eversion can suggest compromise of the medial deltoid ligament. As part of a broader musculoskeletal assessment, the test helps guide diagnosis, injury grading, treatment planning, and decisions about further imaging or rehabilitation.

The video demonstrates the generation of tilted amygdala slices from a mouse brain. The brain is positioned at an angle on the vibratome specimen stage to obtain slices containing the amygdala and axons from the medial prefrontal cortex that form synaptic connections with the amygdala. The brain slices are maintained at the physiological temperature to preserve the tissue for further experiments.

We describe a "gold standard" for evaluating orthostatic tolerance (OT) using tilt testing with combined lower body negative pressure (LBNP). This can be combined with non-invasive evaluations of cardiovascular reflex control. Normal and abnormal responses are defined.

The present protocol describes a generalized and easy-to-implement scheme for tilted single-particle data collection in cryo-EM experiments. Such a procedure is especially useful for obtaining a high-quality EM map for samples suffering from preferential orientation bias due to adherence to the air-water interface.
