Defined Media Formulation

Defined media formulation is the design of a cell culture medium in which each chemical component and its concentration are known, enabling precise control of the cellular environment. By supplying selected nutrients, salts, energy sources, growth factors, and other regulators, the formulation supports cell survival, proliferation, or differentiation while limiting variability from undefined biological materials. In developmental biology, defined media help researchers examine how specific signals influence embryonic cells, stem cells, and developing tissues, including organoid systems. This controlled approach improves experimental reproducibility and supports the study of cell fate decisions, developmental signaling, and tissue formation.

The Culture of Primary Motor and Sensory Neurons in Defined Media on Electrospun Poly-L-lactide Nanofiber Scaffolds

Aligned electrospun fibers direct the growth of neurons in vitro and are a potential component of nerve regeneration scaffolds. We describe a procedure for preparing electrospun fiber substrates and the serum-free culture of primary rat E15 sensory (DRG) and motor neurons. Visualization of neurons by immunocytochemistry is also included.

Freeze-drying is often an easy and convenient way to obtain dry products of viable bacterial cells. An issue of the process is cell survival. We detail here a procedure to investigate how cell survival during freeze-drying is influenced by the properties of the formulation used.
