Calcium Carbonate Measurement

Calcium carbonate measurement is the quantitative determination of CaCO3 in materials such as water, soil, sediment, and geological samples, where it helps characterize alkalinity, buffering capacity, and carbonate mineral content. Common methods add a known amount of acid to react with carbonate, then measure the acid consumed through titration, while instrumental approaches can assess calcium or carbonate directly. In environmental science, these measurements support evaluations of water quality, soil amendment requirements, sediment composition, and carbon cycling. Reliable CaCO3 data also helps researchers assess acidification, mineral dissolution, and the capacity of natural systems to neutralize acidic inputs.

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.
