Heavy Fraction Isolation

Heavy fraction isolation is a separation method used to recover soil organic matter and mineral-associated materials that have greater density than light particulate components. In a density fractionation procedure, a sample is dispersed in a liquid of known density, then settling or centrifugation separates particles according to buoyant density, allowing the heavy fraction to be collected and analyzed. In environmental research, this approach helps distinguish stable carbon pools from more readily decomposed organic matter, assess soil carbon storage, and evaluate how land use, mineralogy, and environmental change influence carbon persistence and nutrient cycling.

We provide a novel strategy to isolate viral replication compartments (RC) from adenovirus (Ad)-infected human cells. This approach represents a cell-free system that can help to elucidate the molecular mechanisms regulating viral genome replication and expression as well as regulation of viral-host interactions established at the RC.

This protocol presents a robust, detailed method to obtain highly pure synaptosomes, synaptic vesicles, and other synaptic fractions from the mouse brain. Crude and fractionated Xenopus egg extracts can be used to provide ingredients for reconstituting cellular processes for morphological and biochemical analysis. Egg lysis and differential centrifugation are used to prepare the crude extract which in turn in used to prepare fractionated extracts and light membrane preparations.
