Mitochondria Enrichment

Mitochondria enrichment is a laboratory process that isolates and concentrates mitochondria from cells or tissues for biochemical, genetic, and functional analysis. The procedure typically uses controlled cell disruption followed by differential centrifugation, in which sequential spinning separates mitochondria from nuclei, membranes, cytosolic components, and other cellular material according to size and density. In genetics, enriched mitochondrial preparations support analysis of mitochondrial DNA, heteroplasmy, genome organization, and mitochondria-associated genetic disorders. By increasing the proportion of mitochondrial material in a sample, this approach can improve detection of mitochondrial variants and enable more focused studies of organelle function, inheritance, and disease mechanisms.

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.

Purification of Mitochondria from Yeast Cells

We describe a rapid and effective method for purification of mitochondria from the yeast Saccharomyces cerevisiae. This method enables the high-yield isolation of pure mitochondria that are essentially free of contamination by other organelles and retain their structural and functional integrity after their purification.

Mitochondria play central roles in the regulation of metabolism and homeostasis. Subtle changes in mitochondrial metabolism that affect organismal physiology could be difficult to detect in whole organism metabolomics studies. Here we describe an isolation method that enhances the detection of subtle metabolic shifts in Drosophila melanogaster.
