Antibody Purification

Antibody purification is the process of isolating immunoglobulins from biological samples or cell culture fluids to obtain preparations suitable for research and clinical applications. The method commonly uses affinity chromatography, in which antibodies bind selectively to immobilized ligands such as protein A, protein G, or a target antigen, while contaminants are removed during washing and the antibodies are recovered by changing pH or ionic strength. Additional steps, including filtration, concentration, or size-exclusion chromatography, can improve purity and stability. In immunology and infection research, purified antibodies support antigen detection, neutralization studies, diagnostics, therapeutic development, and analysis of pathogen-specific immune responses.

2025

This video demonstrates a technique for purifying monoclonal antibodies from the culture supernatant obtained from an antibody-producing cell culture. The process involves using a matrix of crosslinked agarose beads with conjugated Protein A, which immobilizes the antibodies in the supernatant on the beads, allowing for effective separation from contaminants. By applying an elution buffer with low pH, the protein A-antibody interaction is loosened, facilitating the collection of...
