Dextran-coated Beads

Dextran-coated beads are microspheres surrounded by a layer of dextran, a glucose-based polysaccharide, that modifies their surface properties and biological interactions. In immunology, phagocytic cells encounter these particles through surface-mediated binding, followed by actin-dependent engulfment and confinement within phagosomes, allowing bead uptake to be measured under controlled conditions. Researchers use dextran-coated beads to evaluate phagocyte activity, compare cellular responses to particulate materials, and investigate host defense mechanisms relevant to infection. Their standardized composition and visible or detectable cargo also support microscopy, flow cytometry, and quantitative assays of cellular uptake and innate immune function.

This video demonstrates two methods for preparing and placing beads, which have been coated with growth factor, on explants of the developing cerebral cortex. These beads can be used to induce spatially restricted gene expression on developing neural tissue such as forebrain explants. Methods are given for using both Affi-Gel beads and heparin acryllic beads.

The objective of this protocol is to incorporate SDF-1α, a stem cell homing factor, into dextran sulfate-chitosan nanoparticles. The resultant particles are measured for their size and zeta potential, as well as the content, activity, and in vitro release rate of SDF-1α from the nanoparticles.
