Engulfment Reactions

Engulfment reactions are immune-cell processes in which phagocytic cells recognize, surround, and internalize pathogens, dead cells, or other particulate material, helping maintain tissue defense and homeostasis. They begin when surface receptors bind molecular patterns or opsonins on a target, triggering actin-driven extension of the plasma membrane around it and sealing the material inside a phagosome; fusion with lysosomes then exposes the cargo to acidic conditions, hydrolytic enzymes, and antimicrobial molecules. In immunology and infection research, these reactions help explain host-pathogen interactions, microbial clearance, inflammation, and antigen processing, while defects in recognition or intracellular killing can increase susceptibility to infection.

Microglia are the resident immune cells of the central nervous system (CNS) with a high capacity to phagocytose or engulf material in their extracellular environment. Here, a broadly applicable, reliable, and highly quantitative assay for visualizing and measuring microglia-mediated engulfment of synaptic components is described.

Reaction Mechanisms

Chemical reactions often occur in a stepwise fashion, involving two or more distinct reactions taking place in a sequence. A balanced equation indicates the reacting species and the product species, but it reveals no details about how the reaction occurs at the molecular level. The reaction mechanism (or reaction path) provides details regarding the precise, step-by-step process by which a reaction occurs. For instance, the decomposition of ozone appears to follow a mechanism with two steps:
