Gamma Delta T Cells

Gamma delta T cells are a specialized subset of T lymphocytes that bridge innate and adaptive immunity, contributing to early defense against infection and tissue stress. Unlike conventional alpha beta T cells, gamma delta T cells use gamma delta T-cell receptors to detect phosphoantigens, stress-induced molecules, and other targets with limited dependence on classical peptide presentation. Upon activation, they rapidly release cytokines such as interferon-gamma and tumor necrosis factor and can kill infected or transformed cells through cytotoxic mechanisms. In immunology and infection research, these cells help clarify host defense, antimicrobial responses, immune regulation, and connections between inflammation and tissue repair, while their antigen recognition and effector functions support investigation of vaccines and cell-based therapies.

Intravital Visualization of Gamma Delta Intraepithelial Lymphocytes

2025

This video demonstrates the visualization of γδ intraepithelial lymphocytes (IELs) in transgenic mice using confocal microscopy. The surgical procedure involves creating a perforation line and making a longitudinal incision to expose the intestinal mucosa and villi. The transgenic mice express reporter proteins exclusively in γδ IELs, allowing their tracking. Intravital imaging captures the dynamic movement of green fluorescence-labeled γδ IELs along the intestinal epithelial cell's basement...

We describe here an isolation method to obtain human endocervical intraepithelial lymphocytes for the analysis of intraepithelial gamma delta T cells. This protocol can be extended for the purification of endocervical gamma delta T cells by magnetic beads or by cell sorting.

Presented is a protocol for the expansion of Gamma-Delta (γδ) T cell drug product. Lymphocytes are isolated by elutriation and γδ-enriched with zoledronic acid and interleukin-2. Alpha-beta T cells are depleted using a clinical-grade magnetic separation device. The γδ cells are co-cultured with K562-derived, artificial antigen-presenting cells and expanded.

The Delta-to-Delta Circuit

2024

In a delta-delta configuration, the source and the load are connected in a delta manner, forming a closed loop that divides the network into three distinct phases. This configuration makes the phase voltages identical to line voltages. Assuming the sources are in positive sequence, the phase voltages can be expressed directly without having a neutral wire. The phase currents in a delta-connected load are calculated by dividing the phase voltage by the load impedance per phase: In this...

The Y-to-Delta Circuit

A balanced wye-to-delta circuit comprises balanced Y-connected voltage sources and delta-connected loads with no neutral line connection. The initial step in analyzing a wye-to-delta circuit is to assume a positive phase sequence. These phase voltages are then utilized to calculate the line voltages that occur directly across the delta-connected load impedances. Van, Vbn, and Vcn are the phase voltages in wye, and Vab, Vbc, and Vca are the line voltages for a delta circuit. The relation between...
