Glutamine Metabolism

Glutamine metabolism is the network of biochemical reactions that uses glutamine for energy production, biosynthesis, nitrogen transfer, and cellular redox balance. In glutaminolysis, glutaminase converts glutamine to glutamate, which can be further processed to α-ketoglutarate and enter the tricarboxylic acid cycle, while glutamine-derived nitrogen supports nucleotide and amino acid synthesis. These pathways help immune cells meet the metabolic demands of activation, proliferation, and cytokine production. In immunology and infection research, studying glutamine metabolism clarifies how host cells respond to pathogens and how metabolic changes may influence inflammation, immune function, and potential therapeutic strategies.

2014

This article will demonstrate how to monitor glutamine dynamics in live cells using FRET. Genetically encoded sensors allow real-time monitoring of biological molecules at a subcellular resolution. Experimental design, technical details of the experimental settings, and considerations for post-experimental analyses will be discussed for genetically encoded glutamine sensors.

Here we present a step-by-step protocol of the long-length electrostatic repulsion-hydrophilic interaction chromatography-tandem mass spectrometry (LERLIC-MS/MS) method. This is a novel methodology that enables for the first time quantification and characterization of the glutamine and asparagine deamidation isoforms by shotgun proteomics.

FIBS-enabled Noninvasive Metabolic Profiling

A description of how to calibrate Förster Resonance Energy Transfer integrated biological sensors (FIBS) for in situ metabolic profiling is presented. The FIBS can be used to measure intracellular levels of metabolites noninvasively aiding in the development of metabolic models and high throughput screening of bioprocess conditions.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

An Introduction to Cell Metabolism

2023

In cells, critical molecules are either built by joining together individual units like amino acids or nucleotides, or broken down into smaller components. Respectively, the reactions responsible for this are referred to as anabolic and catabolic. These reactions require or produce energy typically in the form of a “high-energy” molecule called ATP. Together, these processes make up “Cell Metabolism,” and are hallmarks of healthy, living cells.JoVE’s introduction to cell metabolism briefly...
